I use a UV-C lamp when I pour agar in my Petri dishes for my mycelium cultures. I let them harden and cool under the light, and then put the lids on when they’ve cooled down. This way I don’t get any contamination or condensation ☺️

I’ve seen in many Master’s Mix recipes calling for 50/50 hardwood to soy hull pellets with 60% water by weight. If I’m doing my math right (and I might not be), it looks like you’re using almost 55% water by weight . Is that correct? If so, do you see much difference in the 5% discrepancy?

75% rate of contamination is far from "pretty good results". And, I wouldnt consider that one plate with the large dot of trich "near clean". Theyre are either clean or contaminated. 3 out of 4 plates tested were most definitely contaminated, not good. If this were my experiment I would consider this "flow hood" an utter failure. With good sterile tek my BBFFU can easily provide over a 90% success rate. It only costs about $70 to build, and takes about 10min to construct with zero tools or mechanical aptitude required. There are inexpensive options out here that do allow for high rates of success without breaking the bank, however the one in this video is not one of them.

I'm Canadian and I'm sorry to say I randomly measure things in pounds and kilograms. There is no rhyme or reason. 9kg and 20lb both make perfect sense in my mind. The one on the right is a spinning disc humidifier. They often get their feed by having a conical underside to the bottom of the disc and the tip of this sits in the water. When the disk is spinning water gets lifted up the cone and thrown out at the periphery of the disc. The problem with this type is that, unless you have a very high rotation speed, the droplets thrown off are not very small - it's difficult to get much output down at 5 microns by this technique. Breaking water into very fine droplets is a high energy job because of the surface tension and the large area you are creating by making the small droplets. This is the kind of fairly useless stuff you learn by doing a PhD in cloud microphysics 😃

i'm very late to the party, but i may have an answer to your final question : Variability in yields. - I used tyndallization like you do in this video until recently, and also had variability in yields. - I bought a real autoclave recently and the variability disappeared. So i went back to tyndallize a few bags until i got one with lower yields to send to the lab, in which they found 2 strains of bacterias, dead. I think the bags aren't always sterilized with this method so the mycelium has to fight foreign bodies, even if it wins, the bacterias or whatever has already "eaten" some ressources and weakened the mycelium (since he has to use ressources to produce antibiotics). The lab showed me the slices they did, and there were obvious zones with no mycelium and different color (where they found the bacterias), you may want to carefully slice your substrates that gave less yields to visually check this.

I will soon do some testing but I guess if you want to increase succes rates grow these truffles I guess you need to isolate the culture, grow it in a completely sealed and sterilized growbag and then use this material to inauculate the trees.

I've watched your one of your videos with the ribbon mixer and I'm 100% sold on this style of concrete mixer, it looks very practical. I think unless scaling up massively just get a second and run them in tandem, mixing one while working with the other substrate, the labour is fine. and if anyone has played No Mans Sky that is exactly what I picture in my head lol
